Muscle creatine kinase isoenzyme expression in adult human brain.
Previous studies have suggested that MM creatine kinase is a muscle-specific protein and is not present in adult brain tissue. We have isolated a protein from human brain with an apparent molecular weight of 43,000 as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis which is identical to the muscle M creatine kinase isoenzyme subunit at all 30 sequenced amino acid residue...
متن کاملMeasurement of brain-specific creatine kinase isoenzyme activity in serum.
We have modified the columti-chromatographic technique of Mercer (1) to produce a sensitive and rapid separation of serum CK2 and CK3, even at normal total serum CK activity (3).12 However, CKj was not separated from CK2. Here, we report a modification of our method such that all three CK isoenzymes are separated by stepwise elution from a mini-column of diethylaminoethyl-cellulose. CK1 has onl...
متن کاملRadioimmunoassay of creatine kinase isoenzymes in human serum: isoenzyme BB.
We describe a sensitive, specific radioimmunoassay for the BB isoenzyme of creatine kinase (CK-BB) in serum. A sequential saturation assay was used to achieve sufficient sensitivity to detect the isoenzyme in 100-microliter serum samples of all healthy persons and patients tested. Bound and free antigen were separated by a second antibody system. Large excesses of purified isoenzyme MM did not ...
متن کاملCreatine kinase isoenzyme patterns in human tissue obtained at surgery.
Chromatography on DEAE-Sephadex A-50 was applied to study the quantitative distribution of creatine kinase isoenzymes in extracts of human tissue obtained during surgery. The results are compared with those determined by an immunological method [Clin. Chim. Acta 58, 223 (1975)]. Conflicting results for some organs as reported by the two methods are probably attributable to postmortem autolysis.
متن کاملStructure of human muscle creatine kinase.
The crystal structure of human muscle creatine kinase has been determined by the molecular-replacement method and refined at 3.5A resolution. The structures of both the monomer and the dimer closely resemble those of the other known structures in the creatine kinase family. Two types of dimers, one with a non-crystallographic twofold symmetry axis and the other with a crystallographic twofold s...
